The sparQ DNA Frag & Library Prep Kit is an integrated enzymatic fragmentation and library preparation kit optimized for construction of high-quality libraries for Illumina NGS platforms. The kit performs combined fragmentation and end-polishing followed by high-efficiency adapter ligation in a simple two-step, single-tube workflow that completes in approximately 2.5 hours and accommodates DNA input amounts from 1 ng to 1 µg. PCR-free workflows are supported from 100 ng of starting material.

Key Features:

  • Format: 96 reactions
  • DNA input range: 1 ng – 1000 ng
  • Total workflow time: ~2.5 hours with minimal hands-on time
  • Workflow: integrated enzymatic fragmentation, polishing (5'-phosphorylation and 3'-dA-tailing), and adapter ligation in a two-step single-tube protocol
  • PCR-free library prep supported from 100 ng input
  • Storage: kit components stored at -25°C to -15°C

Advantages:

  • Generates high-quality sequencing libraries suitable for Illumina platforms with superior coverage uniformity and low duplication rate
  • Tunable and reproducible fragmentation size range enabling control over library insert sizes
  • Fast, streamlined protocol that reduces hands-on time and enables completion of library prep in a single day
  • High-efficiency adapter ligation and high-fidelity amplification options minimize bias and improve library yields
