AccuStart II PCR ToughMix is a 2X concentrated ready-to-use PCR master mix formulated for robust endpoint PCR amplification, especially with challenging or crude DNA templates from environmental, plant, or animal sources. The mix contains an ultra-pure, high-processivity hot-start DNA polymerase combined with high-avidity monoclonal antibodies for room-temperature reaction assembly and specific activation during initial denaturation. It is supplied in a 4000-reaction format for high-throughput laboratory use.

Key Features:

  • Format: 2X concentrated ready-to-use PCR SuperMix
  • Pack size: 4000 × 25 µL reactions (1 × 50 mL)
  • Enzyme: ultrapure modified hot-start Taq DNA polymerase with 5'→3' polymerase and double-strand specific 5'→3' exonuclease activities; no 3'→5' exonuclease activity
  • Room-temperature setup enabled by antibody-based hot-start
  • Includes optional GelTrack electrophoretic mobility dye for direct gel loading

Advantages:

  • Tolerant to common PCR inhibitors found in crude samples, enabling amplification from environmental, plant, and animal extracts
  • Stabilized formulation resists effects of repetitive freeze–thaw and permits convenient room-temperature reaction assembly
  • High-yielding, specific amplification due to ultrapure hot-start polymerase and antibody-mediated activation
  • Optional mobility dye reduces post-PCR handling and lowers risk of cross-contamination during gel electrophoresis
